When I click Acknowledge, the post just dissapears. How do I access it again, how does this now appear to the user, and how is this different than resolving?

The user doesn't see anything different if you acknowledge the discussion. It simple takes the discussion out of your inbox/pending area and just leaves it marked as an open discussion. You can find it again by going to the Open area.

Resolve closes the discussion and marks it as resolved.
